instructions say to begin by drawing 4ml of your tank water & put in test-tube.....i didnt know if you account for the air-bubble or not.

so i'm guessing Example #1 is correct. you always read the black line on the plunger & IGNORE the air-bubble.

That is not the correct syringe for drawing tank water. There should be a 5 ml that was supplied with the kit. That syringe in your picture is 1 ml and used for the test solution
